密室！研究棟のお仕事！

染色体　当時の小生の研究は、はじめ骨髄細胞の染色体検査でありました。主に急性非リンパ性白血病の分析であります。特にＣＭＬ（慢性骨髄性白血病）では有名なＰＨ1染色体がありますが、高精度分染法で処理しますと9番と22番の染色体の転座が確認でき、しかも切断点も同定できます。国際染色体核型表記では、４６,XX,t(9;22)(q34;q11)となります。このように詳細に分析することでその白血病細胞の単一クローンを確認することができます。興味深いことにＣＭＬが急性転化する前に多段階の別の染色体異常（付加的染色体異常）が出現し、より悪性度の高いクローンに変化いたします。実はこのメソードをマスターするために、埼玉癌センターの桜井先生のところに３ヶ月程出向した経験があります。染色体分析のポイントは新鮮な骨髄細胞の同調作業を早く行うことにあります。もちろん骨髄細胞は生きていますからネ！ちなみに自分の染色体を調べてみましたら一応正常核型でありました。
